Abstract:
Through contrast experimen,t the PDY medium which is fit forGanoderma Lucidum. s liqu id fermentat ion and ce llulase production is atta ined, including 200 g potatoes, 20 g glucose, 5 g yeast ex tract and 1 000 mL w ater. The enzyme act iv ity of the cellu lase from Ganoderma Lucidum is measured. The enzyme activities reachs 5. 68 through sa lting out cu lture broth by ( NH4 ) 2SO4 and 6189 through fast prote in liqu id chromatog raphy ( FPLC ), wh ich is respectively 1. 61-fo ld and 1. 96- fold greater than 3. 52 w ithout using thesemethods in the process o f iso lation and purification ofGanoderma Lucidum ce llulase. There are six components. peaks in FPLC spectrum. The spectrum show s that the isolation of the first peak and the second peak is better than that of the o thers and the components of these tw o peaks are h igher. The am ino acid components o fGanoderma Lucidum . s ce llulase are tested, and the characterist ics of cellu lase are analyzed by ultravio let spectrum and infrared spectrum.
