Abstract:
A hybrid peptide gene was designed and synthesized. Its encoding pept ide is constructed from residues 1- 8 o f cecrop in and residues 1- 18 ofmelittin. The came genew as cloned into plasm ids pPIC9K under the contro l ofAOXⅠ and transformed intoP ichia pastoris SMD 1168 stra in by the electroporat ion. By DNA sequenc ing, thewhole sequences o f th is gene are conf irmed correc.t The positive clones w ere identified on YEPD plates contain ing G418 and cultivated in BMMY medium. A fter be ing induced by methano,l a pro tein band can be seen w ith Tric ine-SDS-PAGE. The antibacterial activ ity assay of the product was carried out in a liqu id turb iditymethod and the supernatant o f recombinat ion P ichia pastoris SMD1168 / pPIC9K-came w as exh ib ited to inhibitE. coli DH5α
