Abstract:
To improve the resistance of Chaetomium cupreum against benzamidazole germicide, the benzamidazole resistance gene Benr is transformed into the protoplast of Chaetomium cupreum with the carrier of plasmid pBT6 by polyethy lene glycol( PEG) method. The transformation rate of Chaetomium cupreum is 1. 5×10-5 . Through the analysis of the Southern blot, the gotten transformants are proved to be positive. The transformants have inhibitory effect on Aftrnaria alternate and Rhizoctonia solani and can grow on the potato dextrose agar( PDA) medium containing 60 μg /mL carbendazim. The resistance of the transformants to benzamidazole increases about 100 times compared with that of Chaetomium cupreum wild strains, and can stablize inheritance. The Chaetomium cupreum transformants with resistance against benzamidazole germicide are obtained.
